The genes Notch Accelerates Colon Carcinogenesis

 Corpus Curare Spiritumque

Professor Daniel Louvard (1) (CNRS and director of the Institut Curie Research Center) and his group, in close collaboration with Spyros Artavanis-Tsakonas (2), recently discovered as the gene Notch is the pathogen on the road of cancer of the bowel. The Notch and Wnt signaling pathways play an important role in normal development and homeostasis good.
In mice, the abnormal activation of these signaling pathways increases the number of benign adenomas in the intestine, with a factor of more than twenty compared to the activation of the Wnt signal-alone. In addition, these tumors grow very rapidly in the large intestine, followed by the pathogen in humans are observed. Action for cooperation between the two channels creates a favorable environment for malignant transformation.
These results (published in PNAS) shows that Notch as “accelerator” in the development of colon cancer in humans, which is an essential part of the pathogen. The question is, for a “brake” against them.
Good, corresponding to a surface, the tennis court, is in constant turnover completely renewed itself every five days. This turnover depends on the presence of stem cells and stem cells in the crypts of the intestine villus. Stem cells give rise to stem cells, which are different, successive rounds of division for the different cell types, form, and well. The decisive factor is to find a balance between proliferation and differentiation in Darmepithels.
The Notch and Wnt-signaling pathways are important for maintaining this balance, allowing normal development and organization in the intestine. These two systems of proliferation and differentiation of stem cells in intestinal crypts. The Wnt signal is strictly controlled by the APC (Adenomatosis Polyposis Coli)-Gen under the first phase of two points of carcinogenesis: mutations in one of two copies of this tumor Suppressor-Gen in familial adenomatous polyposis a condition, by the growth of hundreds, even thousands of polyps in the majority of youth. This disease accounts for 1% of cases of colon cancer. The vast majority of sporadic cases of colorectal cancer are also linked to mutations in the APC gene.
The main signal Notch
The Notch gene product as “On / Off” button. When activated, the Notch protein triggers a cascade of intracellular signals essential for the development of all tissues in all multicellular organisms.
Notch is typically as an oncogene and may be a promising therapeutic target.
Professor Daniel Louvard collaboration with Spyros Artavanis-Tsakonas, who has recently shown that the Wnt and into the means of controlling the spread of precursor cells of the cells in the intestine. In animal models, in the aberrantly activated Wnt, Notch activation are dramatically accelerated tumor formation in the intestine, especially in the large intestine. In most mouse models, tumors are rare in the large intestine, as opposed to human patients, the development of multiple adenomas in the colon of mice thus mimics the human pathogens.
This synergy between the Notch and Wnt-genes has been in various animal models, indicating that the mechanism is conserved throughout the development. Louvard professor said that “the Notch gene seems that two points Accelerator carcinogenesis: activation leads to increased cell division and therefore a greater likelihood of change.” However, malignant transformation requires a series genetic events. Aberrant activation of Notch and Wnt-way creates an environment conducive to the accumulation of mutations that ultimately lead to the cancer of the colon. Synergy-Notch and Wnt signals to the early stages of tumourigenesis tumors if called polyps, the stage of screening is so important.
Understanding the mechanisms for education, growth and transformation of these tumors help in the search for new therapeutic targets.
In 2005, France, colon cancer Struck over 37,000 victims and causing 17,000 deaths. The chances of survival of this form of cancer (the third most malignant in the country) are closely related to the stage where the disease is diagnosed.
A mass screening of colorectal cancer was the end of 2008: every two years, all N betwee 50 and 74 are, in a letter, they ask their doctor about colorectal cancer screening and, case by case basis, with a test for the detection of blood in his chair.
